Although probably the reverse of the intended subject the game that came to my mind when I saw the OP was the October 2nd, 1994 Niners vs Eagles game.
The 3-1 49ers were humiliated by the 2-1 Eagles 40-8.
As has been discussed many times (including the NFL Films "Rings" show on the 1994 Niners) this game was the turning point for the Champions-to-be.
Steve Young was pulled (for his own protection?) in the blow-out which precipitated a blow-up of epic proportions. Even Young has stated that he was surprised George Seifert didn't take a swing at him for some of the things

The point being: This was a regular season lost that spurred the 49ers onto an incredible Championship season.
